You choose a random character to play as first. Custom characters aren't allowed.

Then, pick a random cup. Your character has to go in either first, second or third. If your character goes in fourth or under, you cannot play as that character again, at least, until the nuzlocke ends. Custom cups aren't allowed.

This will go on until one character is remaining. In order to finally win this nuzlocke, you must come in first on the final remaining cup with that final remaining character. You can't come in second, nor third, you MUST come in first.
