By Completing Cups in Grand Prix.
Adding onto this, six of the characters in the bottom row are unlocked by completing user-made cups. I’d recommend remakes of the GBA Mushroom Cup since the tracks are pretty easy

to use them there's a thing that should be on the right side of all the normal charecters, click that.
also give them Daisy stats

Also, some of the character are unlocked by getting gold cups in user-made levels.
